Abstract: One of the most important molecular descriptors is lipophilicity, because it indicates a potential biological activity. Lipophilicity of the newly synthesized derivatives of N-substituted-2-phenylacetamide was investigated experimentally using thin-layer chromatography on reversed phase, HPTLC RP18 F254s. As modifiers were used two aprotic (acetonitrile and dioxane) and two protic solvents (ethanol and methanol) and at the same time was varied their volume fraction in the mobile phase. The values of chromatographic retention constant RM0 the compounds were determined. Lipophilicity was determined by computation method, also and the coefficients log P of the studied derivatives were calculated. It was examined the relationship between retention constant RM0 and parameter of lipophilicity log P and good correlation was found. This fact indicates that the retention constant RM0 obtained by thin layer chromatography can be successfully used as a measure of lipophilicity of various organic molecules, and thus to predict their biological activity.
