Abstract: In the blood of patients with chronic misuse of phenacetin-containing analgetics, besides methemoglobin an other pigment with residual absorption of the hemolysate on the wavelength of 632 nm after cyanide addition was a regular finding. By means of differentional spectrophotometry of the hemolysates it was shown that the spectrum of this pigment is identical with the spectrum of hemoglobin derivative produced in vitro with phenylhydrazin and Na2S (sulfhemoglobin). It was found that on 525 nm oxyhemoglobin, sulfhemoglobin and hemiglobincyanid are nearly isobestic, and that on the wavelength of 632 nm the absorptivity of methemoglobin is quite close to that of sulfhemoglobin. These data justify the estimation of met- and sulfhemoglobin separately on the same blood sample through the ratios of the optical densities of the hemolysate on those two wavelengths before and after the addition of cyanide. © 1968 Springer-Verlag.
