dc.description.abstract<p>Klinička primena doksorubicina je i dalje ograničena zbog sistemske toksičnosti ovog antineoplastika, a kao najče&scaron;ći uzrok se navodi oksidativni stres. Cilj ovog rada je bio da se ispita efikasnost nanočestice fullerenola C60(OH)24, kao antioksidativnog agensa u za&scaron;titi od doksorubicinom izazvane nefro-, pulmo- i testikularne toksičnosti. Mužjaci pacova soja Wistar su bili podeljeni u pet grupa i tretirani na sledeći način: kontrolna grupa (I) je primila 0.9% NaCl (1 mL/kg i.p.), druga (II) doksorubicin u dozi od 10 mg/kg i.p., treća (III) 100 mg/kg fulerenola i.p. 30 minuta pre 10 mg/kg doksorubicina i.p., četvrta (IV) 50 mg/kg fulerenola i.p. 30 minuta pre 10 mg/kg doksorubicina i peta (V) grupa je primila samo fulerenol u dozi od 100 mg/kg i.p. Drugog i četrnaestog dana od tretmana, uzeti su bubrezi, testisi i pluća i u homogenatima datih organa mereni su parametri oksidativnog stresa &ndash; lipidna peroksidacija (MDA) i to u grubom homogenatu i parametri antioksidativne odbrane - aktivnost enzima katalaze (CAT), glutation peroksidaze (GSH-Px), glutation reduktaze (GR), glutation-S-transferaze (GST) i superoksid dismutaze (SOD), u citosolu. Pojedinačna visoka doza doksorubicina dovela je do značajnog porasta lipidne peroksidacije i promena u aktivnostima ispitivanih antioksidativnih enzima u odnosu na kontrolnu grupu životinja. Pretretman fulerenolom sprečio je navedene promene u ispitivanim organima jer su gotovo svi ispitivani parametri bili u nivou vrednosti kontrolne, netretirane grupe. Rezultati na&scaron;eg rada podržavaju hipotezu o mogućnosti fulerenola da spreči oksidativni stres i time za&scaron;titi bubrege, testise i pluća od toksičnog delovanja doksorubicina.</p>sr
dc.description.abstract<p>Clinical use of doxorubicin continues to be challenged by its undesirable systematic toxicity, caused mainly by oxidative stress. The aim of this study was to investigate effectiveness of fullerenol C60(OH)24 polyanion nano aggregate and an antioxidant agent, against doxorubicin induced nephro-, testicular and pulmotoxicity. Male Wistar rats were divided into five treatment groups: the control group (I) received 0.9% NaCl (1 mL/kg, i.p.). Groups II, III, IV, and V received single dose of doxorubicin (10 mg/kg i.p.), doxorubicin/fullerenol (100 and 50 mg/kg i.p. of fullerenol as a pretreatment to 10 mg/kg i.p. of doxorubicin) and fullerenol (100 mg/kg i.p.), respectively. Second and fourteenth day after the treatment, organ samples were taken for measurement of the products of lipid peroxidation and activities of superoxide dismutase, catalase, glutathione-peroxidase, -reductase and -transferase. Single high dose of doxorubicin induced a significant increase of lipid peroxidation and alterations of antioxidant enzyme activities, while fullerenol pretreatment prevented the effects of doxorubicin on investigated parameters. Results of this study support our hypothesis of testicular, pulmo- and nephroprotective efficacy of fullerenol in preventing oxidative stress induced by doxorubicin.</p>en
